The Martinos Optics Research facilities consists of multiple separate lab facilities including 1) fiber optic and electronics fabrication and testing, 2) instrumentation system development and testing, 3) small animal studies, 4) optical physics labs with floating tables, and 5) human subject testing. Microscopy

A multi-modal microscopy system, built in house, which includes:

    • 2-Photon Microscope. The microscope uses the galvo-scanning mirrors, 4 PMT detectors covering the 440-710 nm emission spectral range, and it is capable of measuring phosphorescence lifetimes, fluorescence intensity and fluorescence lifetimes.
    • A home built 850 nm Spectral domain Optical Coherence Tomography (OCT) system operating at 22,000 axial scans per second.
    • Laser-Speckle Flowmeter (910 nm wavelength, thermoelectrically-cooled CCD camera: CoolSnap fx).
    • Confocal Microscope (532 nm excitation; photon counting APD from Perkin Elmer).

A prototype 1300 nm Spectral domain OCT system from Thorlabs, Inc. operating at 47,000 axial scans per second.

CCD-based microscopy system for imaging of cortical oxygen tension and blood flow, built in house (180 mJ pulsed laser at 532 nm, Brilliant B, Quantell; TE-cooled 12-bit CCD camera, Imager QE, LaVision; 100 mW laser diode, 830 nm, DL7032-001, Thorlabs).

Near Infrared Diffuse Optical Spectroscopy and Imaging

Two CW-DOT imaging systems (CW6), each with 32 lasers and 32 detectors (manufactured by TechEn).

Four diffuse correlation spectroscopy systems with 760/785/830/850 nm diode pumped solid state lasers, 4 photon counting APD detectors each and high speed timing hardware supporting custom auto-correlation measurements.

IVIS Spectrum CT small animal imaging platform: Bioluminescence, Multispectral fluorescence and spectral unmixing, Cerenkov imaging for optical radiotracer imaging, Low dose and ultrafast microCT. Key features include integrated optical and microCT technology and 3D optical tomography for fluorescence and bioluminescence. The constant horizontal gantry motion and the flat panel detector provide low-dose imaging and automated optical and microCT integration. The stable revolving animal platform table rotates 360° to acquire full 3D data. Multiple animals can be scanned simultaneously while maintaining an average dose per scan at about 13mGy, with a scanning and reconstruction time of less than a minute. Optical and microCT modalities can also operate independently.
